人牙周膜细胞在双氧水处理后纯钛表面的活性

摘要 目的:研究双氧水法处理纯钛表面应用于口腔临床的可行性。方法:倒置显微镜观察细胞原代及传代的生长情况,扫描电镜观察细胞在4种钛片上的附着形态,从形态学、免疫组织化学及细胞增殖、活性方面对比细胞在4种钛片上的异同。结果:人牙周膜细胞在4种钛片上都可以生长及增殖,但其在双氧水处理的钛片上的增殖高于其他3组(P<0.05)。结论:4种处理钛片的方法中,双氧水法更有利于人牙周膜细胞的附着及增殖。细胞在4种钛片上都表现出了碱性磷酸酶(ALP)活性,而且处于稳定后双氧水组的ALP活性最高(P<0.05)。 Objective: To investigate the application possibility of hydrogen peroxide pre-treated titanium in oral clinic. Methods: The proliferation and alkaline phosphatase (ALP) activity of human periodontal ligament (hPDL) cells attached on titanium with four kinds of surface (smooth, rough, etched, hydrogen peroxide treated)were detected. The growth of primary and passaging cells was ob- served with invert microscope, and the morphology of cells was observed with scanning electronic microscope (SEM) ; ALP activity was tested with enzyme-linked immunosorbant assay. The morphology, immunohistochemistry features, proliferation and ALP acyivity of cells on the four kinds of titanium were compared. Results: The hPDL ceils could grow and proliferate on four kinds of titanium. The cell proliferation on titanium treated by hydrogen peroxide was better than those in the other three groups(P 〈0.05). Conclusion: Titanium treated by hydrogen peroxide is more beneficial to attachment and proliferation of the cells than titanium processed by the other three ways. Cells in the four groups all showed ALP activity, and among them, the activity in cells of hydrogen peroxide treated group is the highest(P 〈0.05).
