摘要
目的观察阿托伐他汀对大鼠脑缺血再灌注脑组织MMP-2、MMP-9表达的影响,探讨阿托伐他汀对脑缺血再灌注脑保护作用机制。方法将Wistar大鼠随机分组:正常组(N组)、缺血再灌注组(IR组)、假手术组(NS组)、阿托伐他汀对照组(At组)、阿托伐他汀治疗组(AtT组)、阿托伐他汀预处理组(AtP组)。采用免疫组织化学法检测大鼠脑组织MMP-2、MMP-9表达,用图像分析仪测定光密度值,电镜下分析面密度。结果AtT、AtP组脑组织MMP-2、MMP-9表达较IR组明显降低(P<0.05),较NS、N、At组明显增高(P<0.01);梗死灶体积AtT、AtP组较IR组明显降低(P<0.05),较NS、N、At组明显增高(P<0.01)。结论阿托伐他汀能抑制大鼠脑缺血再灌注后能脑组织MMP-2、MMP-9表达,发挥脑保护作用。
Objective To study the neuroprotective mechanism of atorvastatin against the injury induced by cerebral ischemia-reperfusion. Methods Wistar rats were randomly divided into: normal group (N), ischemia-reperfusion group (IR), sham-operation group (NS), atorvastatin group (At), atorvastatin treatment group (AtT) and atorvastatin preconditioning group (AtP). The rat model of focal cerebral ischemia-reperfusion was established by inserting a piece of nylon thread into the left internal carotid artery. Ischemia of 30 min and reperfusion of 30 min were carried out alternately for 12 h or 24 h respectively. Their expressions of MMP- 2 and MMP-9 in brain tissues were measured by immunohistochemical methods and their optic density value was measured by imaging analysis system. The positive cells for MMP-2 and MMP-9 expressions were counted under optic microscope. Results The expressions of MMP-2 and MMP-9 in AtP and AtT groups were lower than those in IR group ( P 〈 0.05 ), significantly higher than those in NS, N, At groups (P 〈 0.01 ) ; Infarct volume was smaller than that of in IR group ( P 〈 0.05 ), significantly higher than that in NS, N, At groups ( P 〈 0. 01). Conclusion Atorvastatin can downregulate MMP-2 and MMP-9 expressions and restrain infarct volume during focal cerebral ischemia-reperfusion. The treatment with atorvastatin can protect neural function by maintaining the structural integrality of blood brain barrier.
