摘要
目的:比较聚山梨酯-80(吐温-80)、泊洛沙姆188和羟丙基-β-环糊精(HP-β-CD)3种增溶剂对Beagle犬的致敏性。方法:3种增溶剂分别与鱼腥草蒸馏液配伍,采用主动全身过敏试验的方法对Beagle犬静脉给药,观察给药后犬的过敏反应、体温变化、心电图变化以及血常规、肝肾功、血清组胺和总IgE的变化情况。实验同时设鱼腥草蒸馏液单独给药对照组。结果:1%吐温-80组的犬给药后均出现3级以上过敏反应,伴有心率加快,血清组胺水平明显升高。其余3组犬均未出现明显不良反应,且给药前后血清组胺水平没有明显变化。结论:泊洛沙姆188和HP-β-CD与鱼腥草蒸馏液配伍对Beagle犬的致敏性明显低于吐温-80。
Objective ： To compare the sensitization of Beagle dogs to Houttuynia cordata distillation mixed with 3 solubilizers [ Tween-80, hydroxypropyl-β-cyclodextrin （HP-β-CD） and PoIoxamer188 ]. Methods： Houttuynia cordata distillation alone （control） and in combination with Tween-80, HP-β-CD and Poloxamer188 was intravenously injected into the Beagle dogs to induce active systemic anaphylaxis （ASA）. Then the irritability or allergic reaction, the change in temperature and ECG were observed. The routine blood and biochemical variables, serum histamine, and total IgE were determined. Results： The irritability or allergic reactions （ ≥ 3 grade） appeared, and heart rate and serum histamine were increased after intravenous Houttuynia cordata distillation when mixed with Tween-80. No such reactions appeared in other groups. Conclusion： The irritability or allergic reaction induced by intravenous Houttuynia cordata distillation mixed with HP-β-CD and Poloxamer188 are lower than those with Tween-80 in Beagle dogs.
