摘要
目的:分析实验室因素对ELISA法检测HCV抗体结果的影响。方法:选择健康对照组160例和HCV抗体阳性样本组56例,设定反应温度(4℃、22℃、37℃)、温育时间(30、60、90、120 min),以2 ncu/ml弱阳性质控品作为外对照,ELISA法检测HCV抗体。结果:温育(30、60、90、120 min)时检测HCV抗体阳性标本的A值(x^-±s)分别为0.267±0.05、0.387±0.07、0.568±0.08、0.785±0.09,经Newman-Keuel法检验,q分别为11.87、29.77、51.23(均P〈0.01);放4℃冰箱反应30、60、90、120(min)后几乎不显色;在空调室温22℃、水浴箱37℃时,弱阳性质控品HCV抗体的A值(x^-±s)分别为0.203±0.03、0.235±0.04,t=4.70,P〈0.01。不同反应温度和温育时间用ELISA法检测HCV抗体,其A均值经SNK检验,均P〈0.01。轻度溶血(Hb 5 g/L、10 g/L)、轻度乳糜血清(TG1.9-4.0 mmol/L,TC5.3-7.0 mmol/L)HCV抗体阳性标本的A均值分别为0.302、0.321、0.282,与37℃/30 min时的无溶血HCV抗体阳性标本(A均值0.267)比较,t分别为3.044、4.697、1.305,t0.05(56)=5.29,P〉0.05,无统计学意义。结论:要注意避免标本出现严重溶血。注意反应温度、温育时间等实验室因素对酶免疫法的影响。建立标准操作规程,确定符合本实验室实际情况的反应条件。
Objective:To study the three types of common factors which influence the results of antibody to hepatitis C virus by enzyme-linked immunosorbent assay(ELISA).Methods:There were 216 random samples in experimental group and control group,The results were detected using ELISA under the different reaction time(30,60,90,120 min) and temperature(4℃,22℃,37℃),Then ELISA was used to test anti-HCV respectively and the difference between the two groups were compared.Results:A(x^-±s)0.267±0.05,0.387±0.07,0.568±0.08,0.785±0.09,In the experimental group(30,60,90,120 min),P〈0.01).A(x^-±s) 0.203±0.03,0.235±0.04 in control group,t=4.70,P〈0.01;A(0.302,0.321,0.282) in slight lipid and haemolytic serumand,t0.05(56)=5.29,showed no signicant difference(P〉0.05).Conclusion:Research on the influence of anti-hcv by enzyme-linked immunosorbent assay is suitable.
