部分背根切断大鼠脊髓蛋白质组学研究

摘要 目的比较部分背根节切除14d后大鼠脊髓蛋白质与正常组间的表达差异,探讨痛觉传入有关的内在机制。方法取成年SD雄性大鼠16只,随机分为两组。实验组切除双侧L1-L4和L6背根节,保留L5背根节;正常组8只SD大鼠切开暴露背根神经节,但不作切除处理。运用双向电泳和质谱分析技术,比较部分背根节切除14d后大鼠脊髓蛋白质与正常组间蛋白质的肽指纹图和氨基酸序列组成差异。结果部分背根切断14d大鼠脊髓内免疫球蛋白重链V区(Ig heavy chain V region(clone X41-2))表达上调,果糖二磷酸醛缩酶A(Fructose-bisphosphate aldolase A)表达下调。结论发现部分去背根大鼠脊髓存在两个明显差异表达的蛋白质,结果提示,两个差异表达的蛋白质可能与痛觉传入机制有关。 Objective To investigate the importation mechanism of relevant pain sense after partial dorsal root rhizotomy,The differential expression of proteins was compared between the normal and after the partial dorsal root cut 14 days in rats. Methods 16 SD adult male rats were divided into two groups randomly. The L1 -L4 and L6 dorsal root ganglia (DRG) in experimental group (n = 8) were removed,hut L5 was spared. Rats shown up the dorsal root ganglia were designed as control group ( n = 8 ). 2-DE was applied to analyze the difference of the peptide finger print map and amino acid sequence in differential expression of proteins were compared between the normal rats and the partial dorsal root cut ones. Results There was significant difference in the distribution of spinal cord which were heavy chain V region (clone)(41-2 ) of immunoglobulin and fructose-bisphosphate aldolase showed up and down regulations respectively in the experiment group. Conclusion A significant difference in two proteins between the two groups we identified following partial dorsal root rhizotomy in rats. Indicating two proteins may be related to the importation mechanism of relevant pain sense after partial dorsal root cut.
