摘要
操纵生物大分子以在纳米尺度上构造分子建筑,是当今分子操纵领域中很具吸引力的研究主题。本文介绍一种通过改进的“分子梳”技术结合原子力显微镜,进行DNA拉直操纵及成像。先在云母表面制备小分子单层膜,然后将长链DNA转移到该膜上进行操纵。简单讨论了本方法在DNA测序及基因图谱方面的应用。
Manipulating biological macromolecules to form molecular patterns on nanometer scale is an interesting subject with wide prospects.In this paper,by applying “molecular combing” technique and atomic force microscope(AFM),aligned DNA strands on the surface of APS mica system and imaged them.There might be a potential application to sequencing and mapping DNA.
