改组猪IL-2基因与CpG序列对猪伪狂犬病灭活疫苗免疫应答的影响 被引量:5

摘要 将改组的猪白细胞介素-2基因(VRIL2S)与重组CpG(VR1C)质粒经离子交联法制备的壳聚糖纳米颗粒(CNP)包裹后(CNP-VRIL2S-VR1C),肌肉注射给已接种猪伪狂犬病灭活疫苗的免疫猪,接种后第7、14、28、42和56d采集静脉血检测猪的免疫应答变化,并测定血清免疫球蛋白和白细胞介素含量。结果显示,接种组猪血清中IL-2、IL-6、IL-4以及特异性抗体和免疫球蛋白IgG、IgA、IgM的含量显著高于空质粒疫苗对照组,同时外周血液的淋巴细胞和单核细胞数量也较对照组显著增加(P<0.05)。结果表明,CNP-VRIL2S-VR1C能持久显著提高猪对伪狂犬病灭活疫苗的体液免疫和细胞免疫水平,具有被研制为高效安全的分子免疫增强剂的潜力。 The recombinant eukaryotic expression plasmid with the porcine IL-2 gene and CpG motifs were entrapped with chitosan nanoparticles(CNP) prepared by the method of ionic cross linkage,designated as CNP-VRIL2S VR1C,and then were injected intramuscularly into pigs at 0.5 mg plasmid per pig, which were simultaneously co-immunized with the pseudorabies inactivated vaccine. On day 7,14,28,42 and 56 post inoculation,the blood samples were collected from the vein of the experimental pigs to detect the immune parameters. The contents of specific antibody against pseudorabies virus,lgG,IgA and IgM escalated significantly in the sera of the treatment group in comparison with those of the control group(P〈 0.05),and the levels of IL 2,IL-6 and IL-4 were significantly higher in the treatment group than in the control(P-〈0.05). Meanwhile,the counts of lymphocytes and monocytes increased remarkably in the blood of the treatment group compared with the control(P〈0.05). These results indicated that the compound CNP-VRIL2S-VR1C could boost significantly the specific humoral and cellular immunity of pigs to the pseudorabies inactivated vaccine, and promote the immunoprotection for pigs against pseudorabies, which could be developed as a safe and effective promising immunoregulator for development of vaccines against porcine pseudorabies.
