摘要
目的用体视学方法定量研究SD(SpragueDawley)大鼠细菌性重症肺炎模型病理的三维形态结构特点及变化规律。方法SD大鼠随机分为模型组、观察组和对照组(每组分为3小组,前两组每小组8只,对照组每小组4只)。真中,模型组和观察组分剐接种相同浓度不同剂量的肺炎克雷伯菌菌液,对照组接种与模型组剂量相同的生理盐水。分剐于接种后的第2、4、6天分批处死动物。留取肺组织标本,HE染色,每张切片随机取3—4个视野,用Image—Pro图像分析软件分别测试肺实质红细胞体积密度、支气管残余管腔体积密度、肺泡体积密度。结果随着接种时间的延长,模型组、观察组的肺实质红细胞体积密度的变异系数逐渐增大,尤以模型组增大明显;模型组、观察组的支气管残余管腔体积密度变异系数逐渐减小,尤以模型组明显;模型组、观察组肺泡体积密度的变异系数逐渐变小,尤以模型组明显。对照组则无明显变化。结论随着接种时间的延长,模型组、观察组病变逐渐加重,尤以模型组更为明显。在病理体视学上表现为:红细胞在整个肺组织中的体积增加;支气管残余管腔与原始管腔的比积减小;肺泡腔与整个肺实质体积比减小。
Objective To make a quantitative study of the characteristics and the change law of the pathological 3d structure of severe bacteria pneumonia model in Sprague - Dawley (SD) rats with stereological method. Methods The SD rats were randomly divided into 3 groups: model group, observation group and control group; rats in model group and observation group were inoculated with Kleb- siella pneumonia suspension at different doses while rats in control group were inoculated with saline at the same dose of the suspension in model group; the rats in each group were scarified at each time intervals (2d, 4d and 6d after inoculation) respectively, and the lung tissue specimens were collected and were stained with HE ; 3 to 4 visual fields from each section were selected and the volume density of the erythrocyte, the alveolus and the residual lumens of bronchi in the lung parenchyma were analyzed with the Image - Pro image analysis system. Results With the time prolonged, the coefficient of variation (CoV) of volume density of the erythrocyte in the lung parenchyma in model group and observation group kept increasing, especially that in model group ; the CoV of volume density of the alveolus and the residual lumens in model group and observation group kept decreasing, especially that in model group; no obvious changes were found in control group. Conclusions The pathological changes in model group and observation group will get more and more severe with the time of inoculation prolonged, especially those in model group; the pathological stereologieal manifestations are the erythrocyte volume increase in the whole lung tissue, the volume ratio decrease of the residual lumens to the ofigioal lumen of the bronchi and volume ratio decrease of the alveolar space to the whole lung parenchyma.
