摘要
目的研究海蜇头糖蛋白(JF-Ⅲ)体外清除自由基活性及构效关系。方法采用化学发光法测定JF-Ⅲ清除羟自由基和超氧自由基活性,并用化学和酶解的方法,对该糖蛋白清除自由基活性的构效关系进行初步探讨。结果JF-Ⅲ清除自由基活性有显著的剂量依赖关系;JF-Ⅲ中糖部分和蛋白部分同时参与自由基的清除作用;N-连接糖链在清除自由基作用中起主要作用,而O-连接糖链起到较小的作用。结论JF-Ⅲ具有较强的清除自由基活性,其清除自由基能力与三维结构有密切的关系。
Objective To study the free radical scavenging activities and structure-function relationship of glycoprotein (JF-Ⅲ)from jellyfish (R. e) oral-arms. Methods The scavenging activities on superoxide and hydroxyl radicals were estimated by chemiluminescence method, and the structure-function relationships were initial studied by chemical and enzymatic hydrolysis methods. Results The free radical scavenging activities of the glycoprotein had significant dose-dependent relationship. The carbohydrate moiety and the protein moiety were both involved in scavenging activities on free radicals. N-linked sugar of JF-Ⅲ played major roles in free radical scavenging activities, but O-linked sugar played minor roles. Conclusion JF-Ⅲ had strong free radical scavenging activities, which had close relations with its structure.
