摘要
采用原位杂交技术研究地塞米松对大鼠再生肝细胞亚精胺合成酶(spermidine syathase,SPDS)基因表达的影响。结果显示,部分肝切除(partial hepatectomy,PH)后,再生肝细胞mRNA表达量呈现先升高后降低的变化趋势,峰值均出现在PH后9h。PH+去肾上腺使mRNA水平升高,主要表现在PH后6~12h;地塞米松处理组SPDS基因表达量明显下降,并且随着地塞米松剂量升高,mRNA表达水平降低。结果表明,地塞米松对早期再生肝细胞SPDS基因表达具有抑制作用。
The effect of dexamethasone on gene expression of spermidine synthase (SPDS) in regenerating rat hepatocytes was investigated by in situ hybridization. The results showed: in all partial hepatectomy (PH) rats, expressing level of SPDS was gradually rising, then falling with time, and the highest was at 9^th after PH. In the adrenalectomized rats with PH, SPDS mRNA level rose after PH, especially from 6th to 12th h. However, the adrenalectomized rats with PH exposured to examethasone (0.1, 0.2, and 2 mg/kg) , the expressing level of SPDS was reduced with dose-dependent manner. The results suggested that gene expression of SPDS was inhibited by dexamethasone in the regenerating rat hepatoeytes, mainly in early stage of liver regeneration.
