摘要
利用荧光标记法在反相高效液相色谱上对蚕蛹体内痕量前列腺素的分离、鉴定进行了研究,并对不同蛹期蛹体中前列腺素的组成和分布进行了测定。7种前列腺素(PGA1,PGA2,PGB2,PGE1,PGE2,PGF1α,PGF2α)在所选择的实验条件下都能得到很好的分离,色谱柱为C8键合柱(φ6.0 mm ×150 mm),流动相为甲醇/水=70/30,流速为1.5 mL/min,荧光试剂为4-溴甲基-7-甲氧基香豆素;荧光检测波长为Ex=325nm,Em=397nm,最小检测限为 10-10g/mL.
The determination of prostaglandins (PG) in the extract of chrysalis was studied by reversed phase high performance liquid chromatography with fluorescence labeling. The results showed the variation of PG with the life time of chrysalis. The seven kinds of fluores- cence labeled prostaglandins (PGA1, PGA2, PGB2, PGE1, PGE2, PGF1α,PGF2α) could be separated and analyzed with a RP-C8 column (150 mm × 6 mm i. d. ) and methanol/water (70/30 V/V) as mobile phase at a flow rate of 1. 5 mL/min. The 4-bromomethyl-7- methoxycoumarin was used as the fluorescent reagent and the detection wavelength was Ex =325 urn and Em=397 urn. The minimum detectable level was 10-10g/mL.
