摘要
目的:探索胶质瘤免疫治疗的新途径。方法:采用单比实验和联合作用等对比分析,观察抗CD3-抗胶质瘤双抗和相应单抗等多因素细胞毒作用。结果:单用抗体、单用IL-2没有明显的细胞毒作用;LAK细胞+抗体和/或LAK细胞+抗体+IL-2具有明显的细胞毒作用,其中双抗的增强作用明显高于其它各组;源于恶性胶质瘤患者的LAK细胞的细胞毒作用较正常人低,但双抗+IL-2可增强其活性。结论:抗CD3-抗胶质瘤双特异性抗体连同IL-2可将激活的人外周血淋巴细胞特异地介导至胶质瘤细胞后发挥细胞毒作用。
Objective: To explore new strategy of immunotherapy to glioma. Methods: Single and combined comparison experiments were adopted to analyze multiple factors of cytotoxicity of the anti CD 3×antiglioma bispecific antibody and the corresponding monoclonal antibody. Results: No obvious cytotoxicity effects were observed in groups treated by antibody alone nor in groups treated by IL 2 alone. While notable cytotoxicity effects were observed both in groups treated by LAK cell plus antibody and/or in groups treated by LAK cell plus antibody plus IL 2, and the bispecific antibody enhanced cytotoxicity effects a pparently. The cytotoxic activity of LAK cells prepared from patients with malignant glioma was suppressed to some extent, compared with that of health donor, and could be further cooperatively enhanced by BsAb and IL 2. Conclusion: Combined application of anti CD 3×antiglioma bispecific antibody and IL 2 could activate and induce human peripheral blood lymphocytes(hPBLs) to glioma cells, and could mediate and enhance the cytotoxic effects of hPBLs to glioma cells.
