摘要
目的对腹腔注射链脲佐菌素制备糖尿病大鼠模型的方法学进行考察。方法健康雄性Wistar大鼠,按60 mg/kg剂最一次性腹腔注射链脲佐菌素溶液,72 h后,断尾采集空腹静脉血,用葡萄糖氧化酶法测定血糖。结果一次性腹腔注射链脲佐菌素制备糖尿病大鼠模型成功率达到75%,其血糖平均值为(19.464±2.53)mmol/L。模型大鼠血糖值在14d内RSD小于1.29%。结论链脲佐菌素建立糖尿病动物模型过程中动物死亡率低、造模成功率高、稳定性好,是目前较好的一种建立糖尿病模型的药物。
Objective To investigate the method of establishing rat diabetic model by the intraperitoneal injection of streptozotocin. Methods Healthy male Wistar rats were intraperitoneally injected with streptozotocin solution at the dose of 60 mg/kg. After 72 h, the blood was collected from rat vena caudalis, and the blood glucose was determined by glucose oxidase method. Results The achievement ratio of rat model was 75%, and the average blood glucose was (19.46±2.53) mmol/L. The stability was better within 14 d and the RSD value was less than 1.29%. Conclusion Rat diabetic model established by the intraperitoneal injection of streptozotocin has lower mortality, higher achievement ratio and better stability, and streptozotocin is a better drug for inducing rat diabetic model.
