禽流感病毒新疆株A/Duck/XJ/4的M基因的序列分析

摘要 利用RT-PCR技术对禽流感病毒新疆株A/Duck/XJ/4的M基因进行了扩增与克隆,得到了全长核苷酸序列为1 016 bp的M基因,序列分析表明,M基因最大的开放阅读框位于19-1 000碱基;M1蛋白位于19-777碱基,编码252个氨基酸。M2蛋白位于19-44碱基和733-1 000碱基,编码97个氨基酸。同源性分析显示,A/Duck/XJ/4株M基因的核苷酸序列和氨基酸序列与所选H5N1亚型的参考毒株以及所选不同HA亚型的参考毒株均有很高的同源性。 The M gene of A/Duck/XJ/4 strain was amplified by RT-PCR and cloned.The obtained complete sequence of M gene was 1 016 base pairs.Sequence analysis showed that the biggest open reading frame(ORF) was from 19 to 1 000 base pair.M1 protein was located from 19 to 777 base pair and encoded 252 amino acids.M2 protein was located from 19 to 44 base pair and 733~1 000 base pair and encoded 97 amino acids.Homology analysis indicated that M gene of A/Duck/XJ/4 strain,compared with reported H5N1 strains and different HA subtype strains,has high nucleotide Homology and amino acid homology.
