摘要
目的研究超顺磁性氧化铁(superparamgnetic iron oxides, SPIOs)标记乳腺癌干细胞及其生物学特性。方法乳腺癌干细胞分离、鉴定及培养;用Feridex(一种SPIOs)标记乳腺癌干细胞,制备磁标记乳腺癌干细胞;利用MTT、透射电镜和Prus-sian blue染色等方法对磁标记的乳腺癌干细胞生物学特性进行研究。结果在原代培养的乳腺癌细胞中分离出乳腺癌干细胞。Feridex与乳腺癌干细胞共同孵育后,Prussian blue染色及透射电镜均显示乳腺癌干细胞胞浆中含有氧化铁颗粒。随Feridex浓度的增高(15~25μg/mL),Feridex对乳腺癌干细胞存活、繁殖能力的影响差异无统计学意义(P〉0.05)。当Feridex的浓度大于35μg/mL时,影响其存活和繁殖(P〈0.05)。结论本实验利用Feridex作为磁标记探针,对乳腺癌干细胞进行成功磁标记,为进一步利用核磁共振(MRI)对乳腺癌干细胞活体追踪奠定实验基础。
Objective To study the biology characteristic of superparamgnetic iron oxides(SPIOs) labeled breast cancer stem eells(BCSCs), MethYls The culture.and isolation of BCSCs from breast cancer tissue were carried out, Feridex labeled BCSCs were use to be magnetic labeled BCSCs. With Prussian blue staining and transmission electron microscopy(TEM),the cell viability or proliferation biology characteristics were examined. Results BCSCs were found in the culture and dauhgter clones. When BC- SCs were incubated with Feridex,the iron particles appeared within the cells(non-nucleus) and also existed in daughterclones. With the concent ration of Feridex increased(15-25μg/mL), there was no obvious difference of the growth and differentiation of BCSCs (P〉0.05). When the concentration of Feridex exceeded 35μg/mL,there existed an obvious difference(P〈0.05). Conclusion BCSCs are successfully labeled with Feridex that is a kind of SPIOs to be magnetic labeled BCSCs. That is the base of tracking magnetic labeled BCSCs in vivo with MRI.
