组织因子途径抑制物-2在胎膜早破中表达的实验研究

摘要 目的通过检测组织因子途径抑制物-2(TFPI-2)在胎膜早破的胎膜中的变化,探讨其在胎膜早破中的作用。方法实验组:胎膜早破20例,其中未临产行剖宫产者10例(胎膜早破剖宫产组),阴道分娩者10例(胎膜早破阴道分娩组);对照组:未临产剖宫产者10例。采用RT-PCR方法检测上述3组孕妇胎膜TFPI-2 mRNA的表达。Imagetoo图像分析仪分析各组标本相对密度(TFPI-2/-βactin),单因素方差分析各组相对密度差异。结果TFPI-2mRNA在实验组的表达量低于对照组,差异有统计学意义(P<0.05),在胎膜早破剖宫产组中的表达高于胎膜早破阴道分娩组,但差异无统计学意义(P>0.05)。结论胎膜早破中,胎膜的TFPI-2表达减少,胎膜细胞外基质降解增多并导致胎膜破裂。 Objective To investigate the expression and role of tissue factor pathway inhibitor- 2(TFPI-2) in human fetal membrane of premature of fetal membranes(PROM). Methods Amniochorions membranes were prepared from 30 subjects with spontaneous PROM[no labor (NL), n=10 ; normal delivery(ND),n= 10 ]and no labor of intact membranes at term[cesarean section (CS),n= 10]. Messenger ribonucleic acid expression of TFPI-2 were study with reverse transcriptase polymerase chain recction(RT-PCR). Expression density of TFPI-2 in each group was analyzed with Imagetoo analyzer and compared with F test. Results The expression of TFPI-2 in PROM was lower than in intact membranes with significantly difference(P〈0.05). The expression of TFPI-2 in NL was little higher but not significantly different than in ND(P〉0.05). Conclusion The decrease of TFPI-2 could cause increasing of extra-cellular matrix (ECM) degradation and weakening of the membranes lead to rupture.
