控释GDNF联合MSCs源神经元样细胞移植对猴脊髓损伤后前角运动神经元及bcl-2/bax表达的影响 被引量:2

摘要 目的探讨GDNF与MSCs源性神经元样细胞联合移植对猕猴脊髓损伤后前角运动神经元的协同保护作用及机制。方法取12例治疗猕猴脊髓损伤相关的石蜡标本(3组,4例/组),应用光镜统计前角运动神经元数量;应用免疫组化方法及图像分析系统观察bcl-2、bax蛋白表达情况。结果治疗组前角运动神经元数量均明显高于对照组,并且联合移植组数量高于单纯细胞移植组(P<0.01)。联合移植组bcl-2蛋白平均吸光度值高于单纯细胞移植组(P<0.05);Bax蛋白平均吸光度值在两治疗组间表达的差异无统计学意义。结论联合移植组对猕猴脊髓损伤后前角运动神经元的保护作用更佳,可能与GDNF促进前角运动神经元bcl-2蛋白表达增强有关。 Objective To investigate the synergism and mechanisms of transplantaion of controlled-release biomaterial of glial cell lined-derived neurotrophic factor(GDNF) on the motor neurons repair when transplantation with neuron-like cells after spinal cord injury of macaca mulatta. Methods Three groups of spinal cord paraffin specimens of macaca mulatta were collected, every group including 4 cases. Anterior motor neurons were counted and immunohistochemical method and image analysis system were applied in order to observe the expressing of bcl-2 and bax protein. Results Much more neurons could been observed in combined transplantation group and simple cell transplantation group, hardly any neurons could been observed in PBS control group (P 〈 0.01 ). Higher measurement data was observed in the combined transplantation group than the simple cell transplantation group on the average of absorbance of bcl-2 protein (P 〈 0.05 ), but not bax protein. Conclusions Neuron-like cells derived from MSCs transplantion with GDNF produced much more stronger neuroprotective effect than only cells transplantation,which may be relate to the enhanced expressing of bcl-2 protein.
