摘要
采用单因子试验和植物组织培养方法,对4个甘蓝型油菜品种无菌体系的建立以及带芽茎段的增殖和生根进行初步研究。结果表明,4个品种油菜种子最佳的消毒方式为70%酒精消毒30s,再用0.1%氯化汞消毒8~10min;带芽茎段增殖的最佳培养基为MS+6-BA2mg/L+NAA1~2mg/L;最佳生根培养基为MS+NAA2~5mg/L。
Single factor test and plant tissue culture methods were applied to establish the aseptic system of Brassica napus and study the proliferation and rooting of stem with a bud. The best disinfection way of Brassica napus seed was using 70% ethyl alcohol for 30 seconds then using 0.1% HgCl2 for 8 - 10 minutes. MS + 6-BA 2mg/L + NAA 1 - 2mg/L gave the best result for proliferation of Brassica napus stem with a bud and MS + NAA 2 - 5mg/L was best for rooting.
