摘要
目的探讨大鼠三肽基肽酶Ⅰ在正常大鼠组织中的分布及活性。方法用人工合成的Ala-Ala-Phe-MCA底物,采用荧光分光光度计法检测大鼠正常组织中的大鼠三肽基肽酶I的活性大小;用兔抗鼠的三肽基肽酶Ⅰ的抗血清,采用免疫组织化学染色方法观察正常大鼠体内各组织中大鼠三肽基肽酶Ⅰ的分布情况。结果大鼠三肽基肽酶Ⅰ在正常大鼠的各种组织中广泛存在,特别是在肾脏、肝脏及心脏中显著表达。结论大鼠三肽基肽酶Ⅰ广泛存在于大鼠的各种组织中,但表达量及活性有差异。
Objective To investigate the distribution and activity of tripeptidyl peptidase (TPP) I in rat tissues under the normal conditions. Methods TPP I activities of normal rat tissues was detected by spectrofluorophotometer with synthetic Ala-Ala-Phe-MCA. The distribution of TPP I in various rat organs was determined by immunohistochemistry staining with rabbit anti-rat TPP I IgG in antiserum. Results TPP I existed extensively in all kinds of normal rat tissues or organs, with a predominant expression particularly in kidney, liver and heart. Conclusions Rat TPP I extensively exists in all kinds of tissues, but presenting different activities and quantities.
