摘要
目的探讨11β-羟类固醇脱氢酶1型(11β-HSD1)在C57BL/6J小鼠胰腺组织和胰岛β细胞系NIT-1中的表达及其生物学意义。方法以11β-HSD1在肝脏组织中的表达为阳性参照,RT-PCR法和Western blot法检测小鼠胰腺组织中11β-HSD1 mRNA和蛋白表达。细胞免疫化学法检测NIT-1细胞中11β-HSD1的分布与表达,RT-PCR法和Wester nblot法检测NIT-1细胞中11β-HSD1 mRNA和蛋白表达。结果①在正常C57BL/6J小鼠胰腺组织中有11β-HSD1 mRNA和蛋白表达,其表达量均低于肝脏组织(均P<0.01)。②细胞免疫化学染色示NIT-1细胞胞质见棕黄色染色阳性颗粒,同时NIT-1细胞中有11β-HSD1 mRNA和蛋白表达。结论小鼠胰腺组织和胰岛β细胞系NIT-1中有11β-HSD1基因表达,为研究糖皮质激素代谢对胰岛细胞功能的影响以及其它潜在的生理功能,提供了一条研究途径。
Objective To detect the expression of 11β-hydroxysteroid dehydrogenase type 1 (11β-HSD1) in pancreas of C57BL/6J mice and NIT-1 pancreatic beta-cell line established from the transgenic NOD-Lt mouse. Methods The expression of 11β-HSD1 mRNA and protein in pancreas was detected by RT-PCR and Western blot respectively and compared with that in liver. The cellular localization of 11β-HSD1 expression was determined by immunocytochemisty in NIT-1 cells. And the expression of 11β-HSD1 mRNA and protein in NIF-1 cells was detected too.Results The expression of 11β-HSD1 mRNA and protein was detectable in pancreas of C57BL/6J mice, but lower than in liver (P〈0.05). Immunocytochemical staining revealed that there were brown particles in cytoplasm of NIT-1 cells and there was expression of 11β-HSD1 mRNA and protein in NIT-1 cells. Conclusion The pancreatic tissue and NIT-1 cells expressed 11β-HSD1 gene, which supplied a novel means to study glucocorticoid metabolism in pancreatic islets.
