摘要
目的:观察通塞脉片对颈总动脉粥样硬化(AS)组织中金属蛋白酶-2(MMP-2)表达的影响。方法:通过高脂饲料饲养及腹腔注射维生素D_3针建立AS模型,应用通塞脉片对其进行干预,测定血清总胆固醇(TC)、甘油三酯(TG)和低密度脂蛋白胆固醇(LDL-C)含量,及MMP-2的表达。结果:模型组TC、TG、LDL-C含量均高于对照组,差异有非常显著性意义(P<0.01);干预组TC、TG含量也高于对照组(P<0.05)。干预组TC、TG、LDL-C浓度低于模型组,差异均有非常显著性意义(P<0.01)。对照组内膜和中膜仅见散在棕黄色颗粒状阳性染色物质,模型组和干预组可见较多的棕黄色颗粒状阳性染色物质,且模型组阳性染色物质部分融合成片。模型组和干预组MMP-2阳性标记物面积分别为(177.15±4.37)μm^2和(96.45±18.13)μm^2,均明显高于对照组(P<0.05);干预组的表达低于模型组(P<0.05)。结论:通塞脉片干预能降低血脂,抑制MMP-2的表达,有效作用于大鼠颈总动脉粥样硬化斑块而产生治疗效果。
Objective:To observe the effect of Tongsaimai Tablets(TT) on the expression of matrix metalloproteinase-2(MMP-2) in the plaque of carotid Atherosclerosis (AS). Methods:AS rat model was induced by feeding high - fat forage and intraperitoneal injection of vitamin D3,and then was treat- ed with TT.The serum levels of total cholesterol (TC), triglyceride (TG) and low-density lipoprotein (LDL) as well as the expression of MMP-2 in the plaque tissue were detected. Results:Serum levels of TC, TG and LDL were higher in the model group than those in the control group(P<0.01 ), and the TC and TG levels in Tr group were also higher than those in the control group (P<0. 05). TC, TG and LDL levels in TT group were lower than those in the model group(P<0. 01).The positive substance of brown granules in the inti- ma and media was scattered in the control group, and was increased in the model group and TT group.The MMP-2 expression level was (177.15±4.37) in the model group and (96.45±18.13) in TT group, higher than that in the control group (P<0.05), Conclusion: TT is effective for carotid AS in rats by decreasing blood lipid levels and inhibiting MMP-2 expression.
