睫状神经营养因子对脑出血大鼠的神经保护作用

摘要 目的观察睫状神经营养因子(CNTF)对脑出血大鼠的神经保护作用及可能机制。方法实验大鼠随机分为对照组、模型组和睫状神经营养因子治疗组,每组24只,对照组仅给予生理盐水尾壳核注射,后2组采用自体股动脉血注入大鼠尾壳核建立脑出血模型,CNTF治疗组于术后经侧脑室注射CNTF。分别于术后12 h、1d、3d、5d处死取脑,TUNEL法检测神经元凋亡。结果术后各组神经元的凋亡均在3 d时达高峰,CNTF治疗组大鼠脑血肿周围组织神经元凋亡的数量明显低于模型组。结论CNTF对脑出血后损伤神经细胞具有保护作用,可能机制为减少脑出血灶周细胞的凋亡。 Objective To observe the effects of CNTF on cerebral hemorrhage rats and its possible mechanism. Methods Total rats were randomly divided into 3 groups: control group, model group and CNTF therapy group. ICH models were induced in rats of the latter 2 groups with injection of autologous blood into the right caudat, 24 rats in each group. CNTF was injected into lateral cerebral ventricle of rats in CNTF therapy group. Six rats were killed at 12h, ld, 3d, 5d after operation. The apoptosis of cell was detected with TUNEL. Results Compared with model group, there were fewer apoptotic cells in therapy group (P〈0. 05). Conclusion CNTF has protective effects on injured neuron cells, the possible mechanism is that CNTF can reduce neuron apoptosis .
