RNA干扰的机制及多基因表达载体的设计

摘要 RNA干扰(RNAi)是由双链RNA(dsRNA)所引发的靶基因mRNA降解而导致的序列特异性基因沉默,是生物体内抵御病毒感染及由重复序列和突变引起基因组不稳定性的保护机制。随着RNAi研究的越来越深入,能在哺乳动物细胞内持续表达小干扰RNA(siRNAs)的表达载体的建立已成为研究的热点,而同时沉默两个或两个以上基因的表达载体的构建还少有报道。现总结RNAi的机制及几种多基因沉默表达载体的设计方案,期待为RNAi在疾病治疗中的应用提供参考。
