摘要 目的:运用生物信息学方法对新基因FAM92A1功能进行预测分析,为进一步深入研究FAM92A1功能奠定基础。方法:利用多种生物信息学分析软件,分析FAM92A1基因的表达谱、保守性及其编码蛋白的结构和定位等特征,根据结构预测其功能。结果:该基因表达广泛,高度保守,有多个选择性剪接变异体。编码蛋白属于DUF1208保守结构域家族,与猩猩、小鼠、大鼠、鸟、非洲爪蟾、斑马鱼和果蝇的相似性分别为91%、84%、82%、74%、67%、65%、26%。它含有一个卷曲螺旋结构和多个磷酸化位点,无信号肽及跨膜结构。亚细胞定位预测其定位于细胞核。结论:利用生物信息学方法可初步预测FAM92A1为一细胞核内具有调节作用的蛋白质,其在细胞增殖和分化的精确调控方面可能起着重要作用。 Objective To obtain structure and function information of FAM92A1 gene, which will be useful for further research. Methods The EST expression profile and conservation of FAM92A1 gene, the structure and subcellular location of FAM92A1 protein were analyzed by several bioinformaties software. The information obtained from internet was used to predict the function of FAM92A1 gene. Results The expression profile of FAM92A1 analyzed by EST counts showed FAM92A1 was ubiquitously expressed, and homology analysis in NCBI GENE database showed that the FAM92A1 was highly conservative. The FAM92A1 protein belonged to a conserved domain of DUF1208, which contain a coiled coil domain and several phosphorylation sites. Compared Human to Chimpanzee, Mouse, Rats, Gallus gallus, X. laevis, Zebrafish, Drosophila mela- nogaster,the amino acid similarity of FAM92A1 protein is 91% ,84% ,82% ,74% ,67% ,65% ,26% , respectively. There was no signal peptide and trans-membrane structure in FAM92A1 protein. The subeellular location analysis of FAM92A1 showed it was a nuclear protein. Conclusion According to the preliminary inferenee,FAM92A1 is a kind of regulatory protein in nucleus, which may play an important role in the proliferation and differentiation in development.
