川芎嗪对兔失血性休克再灌注中脑的保护效应 被引量:3

摘要 目的:探讨川芎嗪对休克过程脑保护作用的效应及机制。方法:建立兔失血性休克模型,随机分为假手术组(对照组)、失血性休克组(休克组)、休克+回输血+等量NS组(灌注组)和休克+回输血+等量NS+川芎嗪(80 mg/kg)组(灌注+川芎嗪组)4组,每组8只。失血休克组于休克2 h时,灌注组、灌注+川芎嗪组在干预2 h时分别检测脑组织中琥珀酸脱氢酶(SDH)活性、血清和脑组织髓过氧化物酶(MPO)活性及丙二醛(MDA)含量,分别进行组间比较。结果:休克组、灌注组脑组织中SDH活性与对照组比较均降低,差异有统计学意义(P<0.01),与灌注+川芎嗪组比较明显升高,差异有统计学意义(P<0.01)。休克组、灌注组脑组织和血浆中MDA含量、MPO活性与对照组比较均升高,差异有统计学意义(P<0.01),与灌注+川芎嗪组比较明显降低,差异有统计学意义(P<0.01)。结论:川芎嗪对失血性休克代谢障碍及脑组织细胞具有显著的保护作用,可能与通过清除氧自由基、改善线粒体功能,减轻组织休克再灌注损伤有关。 Objective: To study the protective effects of Salidroside on brain metabolism in the hemorrhagic shock through establishing the rabbit model of hemorrhagic shock and reperfusion. Methods: Thirty two rabbits were randomly divided into four groups: the sham operation group (n = 8), the rabbit model of hemorrhagic shock group (n = 8), the rabbit model of hemorrhagic shock with intravenous injection of 0.9% saline group (n =8) and the rabbit model of hemorrhagic shock with intravenous injection of 0.9% saline and salidroside (80 mg/kg) group (n =8). 2 hours after the sham operation, the hemorrhagic shock, the intravenous injection of 0.9% saline and the intravenous injection of 0.9% saline and salidroside (80 mg/kg) were given to each group respectively, the succinate dehydrogenase (SDH) activity in the brain tissue, the myeloperoxidase (MPO) activity and the malondialdehyde (MDA) content in the serum and brain tissue were measured carefully. Results: Compared with the sham operation group, SDH activity in the brain tissue of the hemorrhagic shock group decreased significantly (P 〈0.01), but in the salidroside group, the levels of the enzyme were higher than that in the hemorrhagic shock group and intravenous injection of 0.9% saline group (P〈0.01). MDA and MPO in the brain tissue and serum of the hemorrhagic shock group and intravenous injection of 0.9% saline group were higher than those in hemorrhagic shock group (P 〈0.01), Both of them lowered greatly in the salidroside group (vs. the hemorrhagic shock group and intravenous injection of 0.9% saline group, (P 〈0.01). Conclusion: Salidroside may obviously protect the metabolism disorders and the injury of brain cells from hemorrhagic shock by scavenging oxygen free radicals, improving mitochondrial function, reducing organizations shock and reperfusion injury.
