摘要
目的:通过检测水通道蛋白2(aquaporin 2,AQP2)是否在人胎盘和胎膜组织中表达,探讨AQP2在羊水平衡中的意义。方法:采用逆转录-聚合酶链反应(RT-PCR)和免疫组织化学技术对5例足月正常分娩的胎盘和胎膜组织样本进行AQP2的检测。结果:RT-PCR显示AQP2mRNA水平在胎膜组织中有表达;免疫组化显示AQP2阳性着色主要分布于羊膜上皮细胞的胞膜和平滑绒毛膜滋养细胞。结论:AQP2在人胎膜组织中表达,提示其可能在羊水平衡的调节中发挥作用。
Objective: To detect aquaporin 2 (AQP2) expression in normal human placenta and fetal membranes. Methods: Placenta and fetal membrane (5 cases) were collected from women with intact membranes underwent elective cesarean sections at term. Expression and localization of AQP2 were detected by reverse transcriptase - polymerase chain reaction (RT- PCR ) and immunohistochemistry. Results: AQP2 messenger RNA was found in placenta and fetal membrane by RT - PCR; immunohistochemical localization of AQP2 showed it to be present at epithelial cell of amnion, and cytotrophoblasts of chorion of fetal membranes. Conclusion: AQP2 express in fetal membranes suggests that AQP2 may play an important role in amniotic fluid balance.
