摘要
目的:通过检测水通道蛋白2(aquaporin 2,AQP2)是否在人胎盘和胎膜组织中表达,探讨AQP2在羊水平衡中的意义。方法:采用逆转录-聚合酶链反应(RT-PCR)和免疫组织化学技术对5例足月正常分娩的胎盘和胎膜组织样本进行AQP2的检测。结果:RT-PCR显示AQP2mRNA水平在胎膜组织中有表达;免疫组化显示AQP2阳性着色主要分布于羊膜上皮细胞的胞膜和平滑绒毛膜滋养细胞。结论:AQP2在人胎膜组织中表达,提示其可能在羊水平衡的调节中发挥作用。
Objective： To detect aquaporin 2 （AQP2） expression in normal human placenta and fetal membranes. Methods： Placenta and fetal membrane （5 cases） were collected from women with intact membranes underwent elective cesarean sections at term. Expression and localization of AQP2 were detected by reverse transcriptase - polymerase chain reaction （RT- PCR ） and immunohistochemistry. Results： AQP2 messenger RNA was found in placenta and fetal membrane by RT - PCR; immunohistochemical localization of AQP2 showed it to be present at epithelial cell of amnion, and cytotrophoblasts of chorion of fetal membranes. Conclusion： AQP2 express in fetal membranes suggests that AQP2 may play an important role in amniotic fluid balance.
