摘要
目的:观察胶质细胞源性神经营养因子(GDNF)对局灶性脑缺血大鼠在体海马齿状回脑电活动的影响。方法:制作右侧局灶性脑缺血模型,右侧脑室注射GDNF,通过神经电生理学方法,记录正常组、脑缺血模型组、生理盐水组和GDNF组大鼠局灶性脑缺血90 m in后再灌注14 d海马齿状回的脑电活动,分析脑电频谱。结果:免疫组化检测显示,GDNF组各时间点B r-dU阳性细胞较脑缺血模型组和生理盐水组增多。大鼠脑电图显示,GDNF组较脑缺血模型组和生理盐水组脑电活动恢复明显,组间比较有显著性差异(均P<0.05)。结论:GDNF可改善局灶性脑缺血后的脑损伤,促进激活内源性神经干细胞增殖、分化,改善脑缺血后的行为和脑功能。
Objective To observe the effect of glial cell-lined derived neurotrophic factor(GDNF) on the activity of electroencephalogram (EEG) in the hippocampus dentate gyrus of adult rats after focal cerebral ischemia and to explore the function of GDNF on endogenetic neural stem ceils in the adult rats after focal cerebral ischemia. Methods The right middle cerebral artery of rats were occluded for 90 minutes to establish transient focal cerebral ischemia reperfusion model. GDNF was injected into the left lateral ventricle under the stereo localization. All rats were divided into four groups : normal group, model group, saline group, GDNF group. EEG activity and frequency spectrum of hippocampus dentate gyrus were analyzed in normal group, model group, saline group and GDNF group 14 days after cerebral ischemia reperfusion. Results The recovery of the EEG and frequency spectrum analysis showed rats injected with GDNF were better than those in the model group and the saline group. There were significant difference among groups (P 〈 0.05 ). Conclusion GDNF can enhance the ability of proliferation endogenetic NSCs in the hippocampus dentate gyrus of adult rats and promote the differentiation for NSCs and improve the behavior and function of brain after focal cerebral ischemia.
