摘要
目的探讨小鼠鼻黏膜内T细胞免疫球蛋白黏蛋白分子3(Tim-3)表达水平及其与变应性鼻炎(AR)发病机制的关系。方法BALB/c小鼠20只随机分为变应性鼻炎组(AR组)和正常对照组(N组),每组各10只。AR组:第1,14d腹腔注射鸡卵清蛋白(OVA)100μg加氢氧化铝2mg的生理盐水悬液0.1ml。第21d每侧鼻腔给与1%OVA各10μg,持续7d。N组用生理盐水代替OVA。通过免疫组化和RT-PCR方法检测鼻黏膜内Tim-3表达,同时应用ELISA方法检测血清中IL-4、IL-5及IFN-γ表达,并分析Tim-3与血清中IL-4、IL-5及IFN-γ表达的相关性。结果AR组和N组均有Tim-3表达,AR组表达强度明显高于N组,差异有统计学意义(P<0.05)。Tim-3表达强度与IFN-γ表达量呈负相关,与IL-4、IL-5表达量呈正相关。结论Tim-3在变应性鼻炎的发病过程中可能起重要作用。
Objective To investigate the expression of complement T-cell immunoglobulin- and mucin-domain-containing molecule-3 (Tim- 3 )in mouse nasal mucosa and to reveal its relationship with the pathogenesis of allergic rhinitis. Methods Twenty mice of BALB/c were sensitized intraparitoneally with 100 μg of ovalbumin(OVA) in 2 mg Al(OH)3 on day 1 and day 14. Mice were induced by daily intranasal administration with 10 μg of OVA from day 21 to 28. OVA was replaced with saline in group N. Immunohistochemical staining was used to examine the Tim-3 expression in nasal mucosa. RT-PCR was performed to investigate the level of mRNA expression of complement Tim-3 in nasal mucosa. ELISA was performed to investigate the level of proteinum expression of IL-4, IL-5 and IFN-γ in serum. Results Tim-3 was detected in both groups. Expression of Tim-3 in group AR was higher than that of group N. Pearson correlation analysis demonstrated that the level of Tim-3 was negatively correlated with the level of IFN-γ, but positively correlated with the level of IL-4 and IL-5. Conclusion Tim- 3 may play an important role in the pathogenesis of allergic rhinitis.
