摘要
目的研究HDAC抑制剂MS-275对人肺腺癌A549细胞基因表达谱的影响,探讨MS-275抗肿瘤的分子机制。方法运用制备的包含8064个人类基因的cDNA芯片检测MS-275诱导人肺腺癌A549细胞基因表达谱的变化。结果MS-275(2μmo·lL-1)处理A549细胞16h后,cDNA芯片分析筛选出MS-275对A549细胞的影响基因803个,其中上调基因440个,下调基因363个,其中包括许多与细胞凋亡、细胞分化、DNA修复等相关基因如p21、DR6。结论MS-275通过外源性凋亡信号、内源性凋亡信号和细胞周期阻滞等多种分子机制诱导A549细胞凋亡。
Aim To investigate the effect of HDAC inhibitor MS-275 on the gene expression profile in A549 and explore the potential molecular mechanism of anti-cancer for MS-275. Methods A eDNA microarray containing 8 064 genes was used to detect the gene expression profile induced by MS-275 in A549. Results 440 up-regulated genes and 363 down-regulated genes were obtained in the gene expression profile for A549 treated with 2μLmol·L^-1 MS-275 for 16h. It showed that MS-275 regulated the genes of cell apoptosis, cell differentiation and DNA repair in A549 such as p21, DR6. Conclusion MS-275 induced the apoptosis of A549 cells by various molecular mechanisms such as extrinsic apoptosis pathway, intrinsic apoptosis pathway and cell cycle arrest.
