摘要
目的检测不同病理级别脑胶质细胞瘤中N-myc下游调节基因2(NDRG2)和蛋白激酶B 2(AKT2)蛋白及其磷酸化蛋白的表达水平。方法收集临床上病理级别分别为Ⅱ,Ⅲ,Ⅳ级的胶质细胞瘤组织标本.应用Western-blot方法检测上述蛋白的表达水平,并对其间的相互关系进行分析。结果NDRG2蛋白在脑胶质细胞瘤组织中的含量随着胶质细胞瘤病理级别的升高而降低(P<0.05);在高表达474位丝氨酸磷酸化的AKT2蛋白(P-AKT2-SER474)分子的标本中,两个NDRG2磷酸化的分子多为低表达,相反,在低表达P-AKT2-SER474分子的标本中,两个NDRG2磷酸化的分子多为高表达。结论NDRG2的含量与胶质细胞瘤恶性程度相关;在胶质细胞瘤中,NDRG2的磷酸化不依赖于AKT2的磷酸化,且NDRG2与AKT2的磷酸化作用可能存在竞争的关系。
Objective To investigate the expression levels of N-myc downstream regulated gene 2 (NDRG2) and serine/threonine kinase 132 (AKT2) protein and their phosphorylated forms in different pathological grades of glioma samples. Methods Glioma samples in Ⅱ,Ⅲ, or Ⅳ pathological grade were collected and total protein was extracted respectively. Then Western blot analysis was applied to detect the expression levels of NDRG2 and AKT2 protein and their phosphorylated forms in all samples. All data were analyzed. Results There was clear negative correlation between the expression of NDRG2 protein and the glioma tumor grade (P 〈 0. 05 ). The expression level of two kinds of phosphorylated NDRG2 protein was high in most of samples in which the phosphorylated AKT2 slightly expressed or did not express, and the expression of two kinds of phosphorylated NDRG2 protein was slight in most of samples where the phosphorylated AKT2 highly expressed. Conclusion The expression level of NDRG2 in gliomas correlates with the pathological grade. The phosphorylation of NDRG2 does not depend on the phosphorylation of AKT2 and a competitive mechanism maybe exists in the two processes.
