摘要
[目的]进一步开发利用大花红景天中的多糖。[方法]大花红景天经乙醇脱脂、水提、碱提后,用盐酸中和,乙醇沉淀得粗多糖,经柱色谱纯化得白色红景天多糖,对其进行抗氧化性试验和羟自由基、超氧阴离子自由基的清除试验。[结果]经验证,提取到的多糖不含单糖,为均一多糖。该多糖对猪油有一定的抗氧化作用,对其他油脂也应有一定的抗氧化性作用。羟自由基清除率为50%时,多糖、苯甲酸、甘露醇的浓度分别为208、1751、48μg/ml。超氧阴离子自由基清除率为50%时,多糖和抗坏血酸的浓度分别为1801、18μg/ml。[结论]碱提大花红景天获得的多糖具有较好的抗氧化能力,对超氧阴离子自由基和羟自由基均具有一定的消除作用。
[Objective] The aim of the research was to develop and utilize polysaccharide from Rhodiola crenulata further. [Method]After R. crenulata was degreased by ethanol and extracted by water extraction and alkali extraction, the crude polysaccharide was obtained through neutralizing with hydrochloric acid and ethanol precipitation. White polysaccharide of R. crenulata obtained through purification by column chroro.atography was made for the antioxidation test and the scavenging test of hydroxyl radical and superoxide anion radical. [Result] Through validation, the extracted polysaccharide didn't contain monosaccharide and it was homogeneous polysaccharide. This polysaccharide had certain antioxidation to lard and also had certain antioxida- tion to other lipid. When the scavenging rate of hydroxyl radical was 50%, the concn, of polysaccharide and ascorbic acid were 180 and 118 μg/ml resp. [ Conclusion] Polysaccharide obtained from R. crenulata by alkali method had better antioxidant capacity and had certain eliminating effect on superoxide anion radical and hydroxyl radical.
