摘要
目的建立稳定的乳化系统化学发光法测定脂质过氧化方法。方法对测定系统中乳化剂、测定系统状态、pH值和测定次数(时间)等选择及对测定结果的重复性、线性等方法学试验,考察了乳化系统化学发光法测定脂质过氧化方法的稳定性。结果在pH9.2时,在透明油水分散和乳化两种系统状态脂质过氧化测定结果,他们的变异系数RSD%分别为10.2和26.4;低pH不能启动脂质过氧化,高pH对样品和发光值都有较大影响;在所考察启动脂质过氧化的自由基AAPH(2,2’-偶氮(2-甲基丙基脒)·二盐酸盐)浓度0~125g·L^-1范围内,其浓度与发光值呈良好线性关系,r=0.9944。结论用大豆卵磷脂为乳化剂,系统状态为透明水包油液态微囊,pH9.2,测定能得到较稳定的测定结果。
Aim To improve a chemiluminescence analysis with emulsion for detecting lipid peroxidation. Methods The different emulsifier, state of oil water emulsion, the solution pH and the numbers of the mensuration in measuring system were investigated and selected: Repeatability, stability and linear of experimental results were observed. Results Between two states of oil-water dispersal and emulsion,their chemiluminescence values of coefficient of variation RSD is 10. 2% and 26.4% under pH 9.2 ,The lipid peroxidation can not be started by free radical AAPH at low pH, but at high pH it would considerably effect both samples and chemil In researched congcertration range( AAPH,2,2' -azo-bis-2-amidinopropane-dihydrochloride 0 ~ 125 g · L^-1) ,there was a significant linear correlation between concentration of free radical AAPH and chemiluminescence values ( r = 0:994 4). Conclusion The chemiluminescence analysis with lecithin for detecting lipid peroxidation is repeatable and significantly linear at the state of oil-water dispersal of pH 9.2.
