马尾松实生种子园F1代的RAPD分子鉴别 被引量:1

摘要 为了筛选出鉴定马尾松优良杂种F1代的RAPD特征标记,从DNA水平上区分杂交子代,以从23个马尾松实生种子园F1子代嫩叶中抽提的DNA作为试验材料,采用RAPD分子标记技术检验DNA序列的多态性。从99条引物中筛选出5条适合马尾松F1代RAPD扩增且具有多态性的引物S1026、S1110、S1040、S1180、S1172。利用这5条引物,采用RAPD分子标记技术,对23个F1子代进行了标记基因型的研究,得到了27个标记位点,确立了各样品的标记基因型,并在此基础上进行了马尾松子代的分子分类研究。 In order to choose significant RAPD markers for identification of superior hybrid F1 progeny in Pinus massoniana, and to identify thc progeny at the level of DNA, diversity of DNA sequences.was test by using RAPD molecular marking technique, taking DNA extracted from tender leaves of the 23 F1 progeny in a seedling seed orchard of P. massoniana as experimental materials. Primer S1026,S1110, S1040, S1180, S1172 with diversity were chosen from 99 primers, which were suitable to RAPD amplification of F1 progeny. Marker genotype in 23 F1 progeny was researched, by using the primers and RAPD molecular marking technique, 27 marker loci were obtained, and marker genotype in samples were identified. Molecular taxonomy of the progeny was researched base on that.
