摘要 目的探讨氧化苦参碱对人表皮黑素细胞与角质形成细胞共培养体系酪氨酸酶活性及黑素生成的影响。方法培养人表皮黑素细胞并及人表皮黑素细胞与角质形成细胞共培养体系,利用四甲基偶氮唑蓝(MTT)还原法测定细胞活力,采用酶学方法测定酪氨酸酶活性,475nm比色法测定黑素含量。结果100μmol/L、500μmol/L和1000μmol/L氧化苦参碱对于黑素细胞及共培养细胞酪氨酸酶活性及黑素生成均有较强的剂量相关性抑制作用,500μmol/L及1000μmol/L氧化苦参碱与对照组相比有显著差异(P<0.01)。结论氧化苦参碱对于共培养体系酪氨酸酶活性及黑素生成有较强的剂量相关性抑制作用。 Objective To Investigate the effects of oxymatrine on the acitivity of tyrosinase and melanogenesis in co - cultured model of human melanocytes and keratinocytes. Methods Melanocytes and the co - cultured model of human melano- cytes and keratinocytes were cultured and MTT was used to determine the proliferation of melanocytes and the co - cultures. The activity of tyrosinase and content of melanin were also determined. Results The synthesis of melanin and activity of tyrosinase were clearly suppressed by oxymatrine in a dose - dependent manner at concentrations of 100μM, 500μM and 1000μM in both melanocytes and co - cultures. Statistically significant suppression was observed in oxymatrine at concentrations of 500μmol/L and 1000μmol/L as compared to that of controls (P 〈 0.01 ). Conclusion Oxymatrine exhibits a a dose - dependent inhibitory effects on the synthesis of melanin and the activity of tyrosinase in co - cultures of human melano-cytes and keratinocytes.
