摘要 目的:探讨中西药结合对人胃癌多药耐药细胞SGC-7901/ADR的逆转和诱导凋亡作用.方法:以斑贞1号和/或氟尿嘧啶(5-FU)为阳性对照组,采用MTT法观察斑贞1号与5-FU,川芎嗪(TMP)联合对SGC-7901/ADR细胞的逆转及杀伤作用.采用流式细胞仪(FCM)测定各药物组细胞周期的变化.光镜和电镜观察药物联合前后SGC-7901/ADR细胞形态结构变化.结果:TMP明显提高SGC-7901/ADR细胞对斑贞1号+5-FU的敏感性,逆转倍数为10.39倍.细胞毒作用为:斑贞1号+5-FU+TMP>斑贞1号+5-FU>斑贞1号>5-FU(P<0.01).FCM分析证明3种药物联合与对照组相比可将细胞阻滞在DNA合成前期和静息期-G0/G1期(P<0.05).光镜和电镜下观察到癌细胞体积变小,胞体全面变圆皱缩及典型的凋亡形态学改变.结论:中西药结合对SGC-7901/ADR细胞有较强的逆转和诱导凋亡作用,为当前肿瘤治疗提供了新思路. AIM: To explore the effect of combination of modem medicine and traditional Chinese medicine on the reversal and apoptosis of gastric carcinoma cells SGC-7901/ADR with multidrug resistance. METHODS : BanZhen-1 and/or Fluorouracil(5-FU) was considered as positive controls. MTT assay was used to investigate the effect of combination of BanZhen-1, 5-FUand Tetramethylpyrazine(TMP) on reversal and apoptosis of SGC-7901/ADR in vitro, which was observed by light microscopy. Flow cytometer (FCM) was employed to investigate the cell cycles. The morphological changes of SGC-7901/ADR were observed by transmissional electron microscopy. RESULTS: The sensitivity of SGC-7901/ ADR to BanZhen-1 + 5-FU was enhanced by 300 mg/L TMP. The cytotoxicity of BanZhen-1 + 5-FU also became stronger, with the reversal index(RI) as 10.39. The cytotoxicity effect of BanZhen- 1 + 5-FU + TMP was stronger than that of BanZhen-1 and/or 5-FU (P 〈 0. 01 ). FCM revealed that, with the combitination of BanZhen-1, 5-FU and TMP, cell cycle was blocked at presynthetic phase and ambiguous phase-G0/G1 phase ( P 〈 0.05 ). The light microscopy and transmission electron microscopy revealed that the size of cells exposed to BanZhen-1 + 5-FU + TMP became smaller and the cells strinked, displaying the typical features of apoptosis. CONCLUSION: The combination of modem medicine and traditional Chinese medicine has stronger reversal and apoptosis induction effect on SGC-7901/ADR cells and this may supply a new theoretical basis for the clinical treatment of gastric carcinoma.
