糖尿病皮肤溃疡PDGF及其受体基因表达和蛋白含量的变化 被引量:2

The Expression of PDGF and its Receptor and Changes of Proteins in Dermal Chronic Ulcers in Diabetes

摘要 目的:观察血小板源性生长因子(Platelet derived growth factor,PDGF)两亚基(A,B)和两种受体(α和β)在正常皮肤和溃疡组织中基因表达和蛋白含量的规律及其与糖尿病溃疡形成的关系。方法:用病理学技术检测溃疡组织和正常皮肤的结构特征后,提取8例糖尿病溃疡组织和8例正常皮肤的总RNA后,分离mRNA,用逆转录-多聚酶链反应(RT-PCR)方法检测这4种基因在不同组织中的表达变化,用west-ern blot方法检测这4种蛋白含量的变化规律。结果:在正常皮肤组织内,PDGFR-α和PDGFR-β的基因表达较强,蛋白含量较高;而在溃疡组织中这两种受体的基因表达水平显著降低(P<0.05),蛋白含量分别是正常皮肤的47.3%和35.2%,含量显著减少(P<0.05);而PDGF-A和PDGF-B基因表达水平和蛋白含量在正常皮肤和溃疡组织内差异不显著(P>0.05)。结论:糖尿病溃疡创面难愈性修复可能与PDGFR-α和PDGFR-β基因和蛋白表达水平下降,引起PDGF与受体结合发生障碍相关。 Objective:To investigate the expression of PDGF and its a and β recepters and changes of proteins in normal and ulcer tissues and to find out the relation to ulcer in diabetes. Methods:In 8 cases of diabetes and 8 normals, the structural characteristics of ulcer and skins were detected by pathological technique, the all RNA were extracted and mRNA was isolated, and the expression of four genes was detected by RT - PCR and the changes of four proteins was detected by western blot method. Results:The expression of PDGFR - α and PDGFR-β and the level of proteins in normal tissues was higher than that in ulcers(P 〈 0.05 ). The level of proteins in ulcers was respectively 47.3 % and 35.2 % of normals( P 〈 0.05 ), but the expresiion of PDGFR- A and PDGFR - β genes and the level of proteins between normals and ulcers had no difference( P 〉 0.05). Conelusion. Uncured ulcers in diabetes may be related to lower expression of PDGFR-α and PDGFR-β genes and proteins, which induced in disorder in combination of PDGF and its receptors.
