摘要
目的:延长新鲜猴血球保存期,解决麻疹血凝抑制试验中猴血球来源紧张问题。方法:猴子静脉无菌采血后置阿氏液中,将阿氏液保存的猴血球经500 r/min离心10 min,弃上清收取浓缩猴血球,与等量血球冷冻保存液混合,室温缓慢搅拌15 min后,分装保存于液氮中。结果:将液氮冷冻保存9个月的猴血球与新鲜猴血球比较,其对麻疹血凝素的敏感性和麻疹血凝抑制抗体的测定结果没有统计学差异(u=0.0123,P>0.5)。结论:用液氮冷冻猴血球获得成功,并能替代新鲜猴血球。
Objective:To lengthen the fresh monkey blood cell storage life and to solve the tense problem in the monkey blood cell originate in measles blood congeals inhibition test. Methods: Pick the blood aseptically in the monkey vein, then put in the arab league fluid. Arab league fluid preservation monkey blood cell after 500 r/min centrifugal 10 min, abandons the clear collection concentration monkey blood cell, with the blood cell freezing preservation fluid mix, after in stirring 15 min the room temperature slowly, subpackage preservation in liquid nitrogen. Results:Comparing the monkey blood cells which preserves in the liquid nitrogen freezing nine month - long with the fresh monkey blood cell, the measles haemagglutinin sensitivity and the determination result in measles hemagglutination inhibiting antibody without statistical significance( u = 0. 0123, P 〉 0. 05 ). Conclusion: The liquid nitrogen freezing preservation monkey blood cell are obtained successfully, and they can substitute the fresh monkey blood cell.
