缺氧标记物HIF-1α、GLUT-1在食管鳞癌中的表达及其临床意义 被引量:1

摘要 目的:本文主要探讨HIF-1α、GLUT-1在食管鳞癌中的表达及其临床意义,期望能为食管鳞癌的诊断和靶向治疗提供新的思路和途径。方法:采用SP免疫组织化学染色法检测55例食管鳞癌组织及癌旁不典型增生组织,20例切缘正常组织中HIF-1α、GLUT-1蛋白的表达情况。结果:HIF-1α、GLUT-1在食管鳞癌癌组织中的表达高于正常组织。其表达与患者的性别、年龄无相关性(P>0.05),与食管鳞癌组织的病理分期、分化程度、淋巴结转移有相关性(P<0.05)。HIF-1α、GLUT-1在食管鳞癌中的表达呈正相关。结论:本研究结果提示食管鳞癌组织存在HIF-1α、GLUT-1的过表达,肿瘤的发生、发展中的缺氧过程诱导了HIF-1α的形成,从而促进靶基因GLUT-1的转录,满足恶性肿瘤生长过程中能量代谢糖消耗增加的需要;HIF-1α、GLUT-1可作为判断食管鳞癌预后的参考指标。 Objective. To study the role of HIF-1α and GLUT-1 in esophageal carcinoma and their relationship. Methods. None of the 55 cases involved in this study had received radiotherapy and chemotherapy. The expression of HIF-1α and GLUT-1 in esophageal carcinoma was detected by SP immunohistochemistry. Results. In the 55 cases of esophageal carcinoma, the positive rate of HIF-1α and GLUT-1 expression was respectively significantly higher than that in normal tissue. No differences were discovered in the expression of HIF-1α and GLUT-1 protein when the age and the gender were considered. However, the expression of HIF-1α and GLUT-1 are correlated to clinic TNM stage, histological grade and distant metastasis of esophageal carcinoma. There were certain positive correlations between HIF-1α and GLUT-1. Conclusion. HIF-1α and GLUT- 1 may be considered as a reference criterion in evaluating esophageal carcinoma.
