摘要 目的检测DNA聚合酶ι(Polymerase iota,Polι)在尿路上皮癌细胞株及肿瘤组织的表达,探讨Polι在尿路上皮癌组织表达的意义。方法培养尿路上皮癌细胞株BIU87、T24,反转录聚合酶链式反应(Reverse transciptase-polymer-ase chain reaction,RT-PCR)检测Polι在尿路上皮癌细胞株的表达;收集人膀胱黏膜正常组织及临床膀胱尿路上皮癌、肾盂尿路上皮癌组织标本,检测组织标本中Polι的表达。结果Polι在尿路上皮癌细胞株有丰富表达,显著高于膀胱正常黏膜组织;Polι在膀胱尿路上皮癌及肾盂癌组织的表达显著高于膀胱正常黏膜组织的表达(P<0.01),并与尿路上皮癌的分级有关。结论Polι在尿路上皮癌组织的高表达可能与尿路上皮癌的发生、发展有关。 Objective To investigate the significance of DNA polymerase iota (Polt) expression is transitional cell carcinoma. Methods RT-PCR was applied to detect the expression of Polt in BIU87 and T24 cell lines, and in tissue samples of transitional cell carcinoma from bladder carcinoma and renal pelvic carcinoma. Results Expression of Polt was slight in normal bladder mucous membrane but significantly elevated in transitional cell carcinoma cell lines. Compared with Polt expression in normal bladder mucous membrane, the expression was significantly increased in transitional cell carcinoma tissues ( P 〈 0.01 ) and also associated with the grade of transitional cell carcinoma. Conclusion The significantly increased expression of polymerase iota may be associated with the genesis and development of transitional cell carcinoma.
