木黄酮对体外培养的HSC-T6细胞基质金属蛋白酶抑制因子1基因表达的影响 被引量:1

摘要 目的以雌二醇(E2)作为阳性对照,观察植物雌激素木黄酮(GST)对体外培养的HSC-T6细胞基质金属蛋白酶抑制因子1(TIMP-1)基因表达的影响。方法E20.1μmol/L浓度作为阳性对照,GST 0.5、5、50μmol/L浓度分别作用于HSC-T6细胞36 h,以RT-PCR方法测定其对HSC-T6细胞TIMP-1基因表达的影响。结果E2浓度组和GST各浓度组均对HSC-T6细胞TIMP-1基因表达有明显的抑制作用,其中GST各浓度组作用呈一定剂量相关性。结论GST各浓度组均可抑制HSC-T6细胞TIMP-1基因表达水平,从而促进胶原降解,可能是其抗肝纤维化的作用机制之一。 Objective By using estradiol (E2) as positive control to observe the effects of genistein (GST) on the matrix metalloproteinase inhibitor 1 mRNA levels of HSC-T6 cell in vitro. Methods HSC-T6 cells were exposed to different concentrations of E2 0.1μmol/L and GST 0.5-- 50 μmol/L for 36 hours. The TIMP-1 mRNA levels were measured by the quantitative reverse-transcription polymerase chain reaction (RT-PCR). Results The TIMP-1 mRNA levels of HSC-T6 cells at different concentrations of E2 and GST were lower than those of the normal control (P〈0.01). We found that the more concentration of GST group the more inhibitory effects of the TIMP-1 mRNA levels. Oonciusion The antifibrotic mechanism of GST is partly due to its downregulation on TIMP-1 mRNA levels of HSC-T6 cells.
