摘要
①目的探讨ＭＴＴ比色法在化疗药物敏感试验中的适宜条件。②方法用膀胱癌细胞株（ＢＩＵ－８７）为亲本细胞，对影响ＭＴＴ比色法实验条件进行了研究。③结果ＭＴＴ比色法较好的反映细胞的增殖程度。细胞数量、ＭＴＴ浓度、残留培养液均可影响实验结果。ＢＩＵ－８７细胞株对不同化疗药物的反应性不同。④结论ＭＴＴ比色法受多种因素影响。选定适宜实验条件，可为临床化疗药物筛选提供参考。
Objective To investigate the methodology of tetrazolium based colorimetric assay(MTT assay) in chemosensitivity test of chemotherapy. Methods Various factors involved in the MTT assay was studied by using a human bladder tumor cell line——BIU 87. Results MTT colorimetric assay could be used to measure cell survival and chemosensitivity. The assay was influenced by both MTT concentration and the medium left in the culture plates. The chemosensitivity of BIU 87 to several chemotherapeutic drugs were different. Conclusion The result of MTT assay is affected by several factors. MTT assay may be used for clinical reference in selecting drugs with suitable condition.
