摘要
目的:研究肝纤维化过程中胶原成分的变化规律.方法:制作四氯化碳肝纤维化模型,测定肝组织羟脯氨酸含量,用VG、RF染色显示Ⅰ、Ⅲ型胶原并进行计算机图像分析,放免法测定肝匀浆及血清Ⅲ型前胶原(PCⅢ)水平,应用上述指标反映肝纤维化过程中胶原的变化规律.结果:随实验时间延长,肝内胶原含量逐渐增加,肝纤维化早、中期以Ⅲ型胶原增生为主,肝纤维化后期及肝硬化期以Ⅰ期胶原增生为主.血清PCⅢ与肝内胶原变化基本同步.结论:肝内胶原增加是肝纤维化过程的病理基础,血清PCⅢ可作为反映肝纤维的指标.
Aims:To research the changes of collagen during the period of development of liver fibrosis induced by CCL4 in rats. Methods: The contents of hepatic hydroxyproline, volume density of collagen type I and type III, the levels of procollagen type III (PCIII) in sera and liver homogenate were measured.The histological findings were also observed by light and electron microscopy.Results:The quantity of collagen in hepatic parenchyma increased gradually. In the early and middle stages of fibrosis, the amounts of type III collagen significantly increased in liver, however, in the final fibrotic stage and cirrhotic stage, type I collagen increased dominately. Conclusions: These results indicated that increases of collagen in liver were a basic pathological process in liver fibrosis and the levels of PCIII in sera might be useful parameter to diagnose the liver fibro-
