由家蚕蛹─BmNPV系统建立环境诱变剂检测分析模型的初步研究 被引量:6

摘要 报道了9─氨基吖啶(9-AA)、甲基磺酸乙酯(EMS)和丝裂毒素C(MMC)3种强诱变剂在新设计的家蚕蛹─BmNPV系统致突变检测分析模型中得到的初步结果。9-AA和EMS以非中毒剂量分别与BmNPV混合注射于蚕蛹体腔后,2种诱变剂处理组蚕蛹发病时间比只注射病毒对照组延迟2~3d,随剂量增高,发病死亡率下降,蚕蛹生存率上升,存在明显的剂量效应。镜检3种诱变剂处理组蛹的血淋巴,发现部分病蛹中出现5%~20%的异常形态多角体,说明蛹体内部分BmNPV的基因可能被诱变损伤。试验结果符合模型分析第一阶段的预期结果。 This paper reports a newly designed model to detect environmental mutations with silkworm-baculovirus system and the mutation results of ethyl methanesulfonate (EMS), 9-aminoacridine (9-AA) and mitomycine (MMC) detected in the model. 9-AA and EMS of non-toxic dosage were mixed with wild Bombyx mori nuclear polyhedrosis virus(BmNPV) respectively, then injected into the silkworm pupae separately. The infection period in the pupae treated with the above two mutagens were delayed 2-3 days than the check group with only BmNPV,and the higher the dosage of the mutagens was ,the lower the mortality and the higher the survival rate would be. About 5 %~20% of polyhedra were found changed morphologically in the hemolymph of some diseased pupae by microscope and electronic microscope. It implies that the polyhedrin gene of BmNPV may be mutated by these mutagens. The results were in accordanLe with the expected ones in the first step of analysis with the model.
