摘要 目的:用微核试验初步评价芦荟提取物的遗传毒性。方法:健康昆明种小鼠分别灌胃芦荟提取物样品1.1,2.2,4.4 g·kg^(-1),qd,连续4 d,同时设阴性(注射用水,腹腔注射)和阳性对照组(环磷酰胺,腹腔注射),每组10只,雌雄各半。每只小鼠的骨髓涂片计数1000个嗜多染红细胞(PCE),记录含微核的嗜多染红细胞(MNPCE)数,计算微核率。结果:芦荟提取物低、中、高3个剂量组的微核率依次为(13.60±3.50)‰,(20.60±3.94)‰和(17.30±4.16)‰,环磷酰胺阳性对照组的微核率为(18.40±1.07)‰。与阴性对照组的微核率[(4.40±0.84)‰]比较,芦荟提取物低、中、高3个剂量组和环磷酰胺组均显著升高(P<0.01),中剂量组的微核率最高,还显著高于阳性对照组(P<0.05)。结论:芦荟提取物能明显诱导小鼠骨髓细胞微核率增加。 Objective:To primarily evaluate the genotoxicity of the Aloe extract by the micronucleus test. Methods: In healthy Kunming mice, the Aloe extract was intragastricaily administrated at doses of 1.1 , 2.2 and 4.4 g· kg-1 , once a day for 4 days. Injection water and cyclophosphamide were respectively used as the negative and positive controis, and 10 mice (5 males and 5 females) were used in each group. For each mouse, the number of micronucleated polychromatic erythrocytes (MNPCEs) in 1 000 polychromatic erythrocytes (PCEs) was counted. Results: The frequency of MNPCEs was ( 13.60 ±3.50)%v, (20.60 ±3.94)%0 and ( 17.30 ±4.16)%o, respectively, in Aloe 1.1, 2.2 and 4.4 g·kg-1 groups, and (4.40 ±0.84)%0 and (18.40 ±1.07)%o in the negative and the positive control groups. The Aloe extract and cyclophosphamide significantly increased the frequencies of MNPCEs ( P 〈 0.01 ). The frequency of MNPCEs in Aloe 2.2 g·kg-1 group was the highest, which was significantly higher than that of the positive control group (P 〈 0.05 ). Conclusion : The Aloe extract significantly increases the frequency of micronucleus in polychromatic erythrocytes in mouse bone marrow.
