摘要
为了直接可视化观察焦亚硫酸钠(SM)与红细胞作用后形态上的变化以及对细胞膜分子形貌的影响,利用纳米分辨率水平的原子力显微镜对经不同SM浓度的红细胞表面进行扫描观察。结果表明:与正常红细胞相比,经SM作用后的红细胞的高度明显降低,凹陷部分更加明显;对红细胞上小范围扫描成像的结果显示,受损后的红细胞膜表面结构发生了变化,膜表面颗粒排列的特征依然存在,但颗粒之间开始产生连接并团聚;SM具有生物毒性,能损伤红细胞膜,改变细胞膜的微结构,引起红细胞的溶血作用。
To observe damaging effect of sodium metabisulfite (SM) on the natural molecular morphology and cell membrane surface ultrastructure changes of red blood cells (RBC) directly, atomic force microscope (AFM) with nanometer resolution was used to scan the RBC after being treated with SM at different concentration. The experiment suggests that in comparison with normal RBC, after being treated with SM, the thickness of RBC was reduced and the concave became more clearly. Small size of RBC membrane was scanned. The images show that there ate some changes in the ultrastructure of damaged RBC membrane. The regularly arranging characteristic of particles disappears and the membrane surface becomes rough. It is concluded that SM have the biotoxicity and can damage RBC and change the ultrastructure of RBG membrane.
