摘要
目的:探讨地塞米松诱导小鼠腭裂动物模型的建立方法。方法:采用一次性腹腔注射地塞米松的方法,建立腭裂模型鼠。取孕12.5d、13.5d、14.5d、15.5d、16.5d胚胎头部,石蜡包埋,6μm切片,HE染色,观察腭发育过程腭突的变化。结果:妊娠第11.5d一次性给予地塞米松(Dex)1mL(用量为50mg/Kg)可诱发小鼠产生腭裂。模型组腭突体积瘦小,不能在中线区接触融合,以致不能与腭突上方的鼻中隔接触融合,胎鼠出现腭裂。结论:妊娠第11.5d一次性给予小鼠足量地塞米松,可成功建立腭裂动物模型。为研究腭裂形成原因及分子机制提供了一个较好的模型。
Objective: To investigate the method of constructing the model of cleft palate induced by dexamethasone(DEX). Methods: The cleft palate mouse embryo model was got by injecting DEX intraperitoneally into Kunming mice. The heads of the mouse's embryo were got from GD12.5 to GD16.5 ,then fixed with Bouin's solution,embedded by paraffin,sectioned 6μm and HE stained. The changes of palatine process were observed during the development course of cleft. Results: Cleft palate at GD 11.5 could be induced by injecting DEX at the dose of 50mg/kg once. The palatine process and anasl septum,which could not contact and merge each other,was thin and small in the model group so as to develope palate cleft on the embryo. Conclusion: Injecting enough dose DEX once at GDll. 5 can induce the cleft palate mouse embryo model successfully. It is a perfect model for studying the reasons and mechanism of cleft palate forming.
