摘要 质粒的分离纯化在分子生物学实际工作中占有重要地位,本文依据DNA在高盐条件下吸附于氧化硅介质,低盐条件下脱附的原理,采用包裹有氧化硅的磁性纳米粒子,粒径为20nm左右,在外磁场作用下提取细菌质粒DNA,操作简便,在20rain内完成.从1~3mL过夜培养的大肠杆菌LB培养液中可纯化得到1~30μg的高纯度质粒DNA,该质粒DNA可直接用于分子生物学下游操作. High-quality purified plasmid DNA is very important for experimentM studies of molecular biology and biomolecular analysis. This paper describes the development of a simple and reliable method for the rapid isolation of plasmid DNA from crude cell lysates with silica-coated magnetic nanoparticles, which are synthesized with an average particle size of 20 nm. This approach for plasmid DNA isolation has many advantages compared with the conventional plasmid purification process. The plasmid obtained by this approach retains its biological activity that is suitable for restriction enzyme digestion and cell transformation.
