银杏叶提取物预防大鼠肝纤维化的实验研究 被引量:1

摘要 目的研究银杏叶提取物(EGb)预防大鼠肝内纤维组织形成的动态变化。方法58只SD大鼠随机分为正常对照组(10只)、模型组(16只)、EGb防治组(16只)和秋水仙碱组(16只)。用四氯化碳皮下注射法建立慢性肝损伤模型,分别用生理盐水或药物灌胃治疗,于第4、8周随机各处死8只大鼠,动态观察EGb对慢性肝损伤大鼠的肝功能、肝纤维化、肝病理组织学及脂质过氧化指标的影响。结果EGb防治组肝功能指标ALT、AST、白蛋白(ALB)、AKP明显改善(P<0.05或P<0.01);肝纤维化血清学指标透明质酸(HA)、层黏连蛋白(LN)含量明显低于模型组(P<0.05或P<0.01);肝组织中丙二醛(MDA)含量较模型组降低,过氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性明显高于模型组(P<0.05或P<0.01);VG染色示肝组织纤维化程度较模型组明显减轻(P<0.05);EGb综合效果优于秋水仙碱。结论EGb对慢性肝损伤有良好的保护作用,能显著减轻纤维化。抗氧化作用可能是其主要的作用机制之一。 Objective To observe the effect of extract of ginkgobiloba (EGb) on hepatic fibrosis in rats. Methods Fifty-eight SD rats were divided into blank contrul group (10), model group (16), EGb group (16)and colchicina group (16)in random. Chronic liver damage model was established by hypodermic injection of carbon tetrachloride, the saline and drugs were given by intragastric administration. Eight rats were killed on 4th and 8th week respectively. The effects of EGb on liver function, hepatic fibrosis, liver histopathology and lipid peroxidation were observed. Results ALT, AST, ALb and AKP of the rats in EGb group were better than that of the model group( P〈0.05orP〈0.01); content of MDA in tissue of liver in EGb group is lower,SOD and GSH-Px were higher( P 〈0. 05 or P 〈0. 01 ) than that of model group;compared with model group, staining of VG showed liver tissue fibrosis of EGb group was decreased ( P 〈 0. 05 ) ; in general, EGb is better than col- chicin. Conclusions EGb could protect liver from chronic damage in rats and obviously decrease hepatic fibrosis. It is possible that antioxidation of EGb plays a key role in its mechanism.
