微波辐射对大鼠瞬膜条件反射的影响

摘要 目的建立大鼠瞬膜条件反射的实验模型,观察微波辐射对大鼠瞬膜条件反射的影响。方法10只二级雄性Wistar大鼠,采用1000Hz的正弦波纯音(条件刺激)和压强为80g/cm2的束状气流(非条件刺激)同时作用于左眼角膜,训练其形成瞬膜条件反射,评价该实验方法的可行性。选择已经建立牢固瞬膜条件反射的大鼠20只,随机分为对照组和微波辐射组(包括辐射后0h、3h、24h和72h等四个时相点),辐射组给予平均功率密度90mW/cm2的S波段微波持续辐射20min,测定辐射后大鼠的即刻肛温并计算比吸收率值;检测辐射后各时相点大鼠瞬膜条件反射的变化。结果10只大鼠通过1周的训练后均能够形成牢固的瞬膜条件反射(平均习得率为90.50%),训练结束后1周内瞬膜反射可较好地保留(平均保留率为84.50%)。90mW/cm2微波辐射后即刻大鼠肛温升高2.89℃,SAR值为34.99kJ/kg。瞬膜条件反射在辐射后0h显著降低,3h后基本恢复至正常水平。结论本实验中大鼠瞬膜条件反射模型的制作方法较简单,可操作性强,实验结果较稳定。90mW/cm2微波辐射20min能使大鼠机体产生明显热效应,并损害大鼠的瞬膜条件反射。 Objective To train rat to form eye blink conditioning, and to investigate the effect of microwave irradiation on eye blink conditioning of rat. Methods Ten male Wistar rats accepted both sinusoidal wave simple tone(frequency is 1000Hz) as conditioned stimulus and sarciniform air current( pressure is 80g/cm^2) as unconditioned stimulus at cornea of left eye to form eye blink conditioning, in order to evaluate this method was feasible whether or not. Twenty male Wistar rats(having steady eye blink conditioning)were divided randomly into control and microwave irradiation group( including 0, 3, 24 and 72 hours subgroups after irradiation) , rabbits accepted 90 mW/cm^2 S frequency range microwave irradiation for 20 minutes, rectal temperature were detected immediately after irradiation and specific absorbtion rate(SAR)value were calculated, eye blink conditioning were detected immediately after irradiation. Results Ten rats established steady eye blink conditioning after seven days training(average positive rate was 90.50% ) , and conditioning remained very well after training for seven days(average positive rate was 84.50% ), Rectal temperature of rats increased 2.89 ℃after microwave irradiation, SAR value is 34.99 kJ/kg; Eye blink conditioning decreased significantly after irradiation. Conclusion The method of training eye blink conditioning is simple and feasible. After 90 mW/cm^2 microwave irradiation for 20 min, rats body occured heating effect obviously, and rats eye blink conditioning injured significantly.
