用于祛除皱纹的真皮成纤维细胞体外培养的研究 被引量:2

摘要 目的探讨体外快速培养真皮成纤维细胞的方法。方法取SD乳鼠背部一块皮肤,运用组织块培养方法将皮肤剪成1mm×2mm的组织块,用胰蛋白酶消化成细胞悬液,培养真皮成纤维细胞,然后观察成纤维细胞的生长情况。结果SD乳鼠真皮组织块法培养72h后呈现单细胞生长,SD乳鼠真皮悬液法48h即可进入倍增生长期,成人真皮组织块法从细胞开始生长7—10d后传代1次。结论建立了一种新的、适宜于大鼠和人皮肤成纤维细胞的培养方法,能快速培养出高纯度的真皮成纤维细胞,以用于生物除皱。 Objective To study the fast method to culture dermal fibroblast in vitro . Methods Cultured dermal frbroblast in vitro with tissue pieces and enzymatic digestion, then observed the growth of cells. Results The cultured cells with tissue pieces and enzymatic digestion were shuttle shape, which showed the cells were pure fibroblasts. A lot of cells can be obtained fast. Conclusion We found a new method which is suitable to culture fibroblast of mice and human. They can be used for rhytidoplasty because of high purity and number.
